摘要
采用6-氨基喹啉-N-(羥基琥珀酰亞胺基)氨基甲酸酯(AQC)為柱前衍生化試劑,建立了AQC柱前衍生化反相高效液相色譜法用于測(cè)定大蒜中的蒜氨酸及其有關(guān)物質(zhì)的含量。

該衍生化反應(yīng)可瞬間完成,衍生化產(chǎn)物穩(wěn)定。采用的色譜條件為:KromasilC18柱(250mm×4.6mm,5μm),流動(dòng)相A為0.1%乙酸銨(含0.03%乙酸),流動(dòng)相B為水-乙腈(體積比為40∶60),線(xiàn)性梯度洗脫,流速為1.0mL/min,檢測(cè)波長(zhǎng)為248nm。蒜氨酸在1.1719~1500μg/mL范圍內(nèi)線(xiàn)性關(guān)系良好(r=0.9998),日內(nèi)、日間測(cè)定的精密度良好(相對(duì)標(biāo)準(zhǔn)偏差(RSD)為1.8%,n=5),添加回收率為99.1%(RSD=1.9%,n=5),最小檢出濃度為0.15μg/mL。該方法準(zhǔn)確、方便、快速。
